Traditionally, ion channels are classified according to the mechanism that controls the opening and closing transition (gating) and the type of ions that can pass through the channel (selectivity). These functional characteristics, as well as the control of expression and positioning, determine the effector activity of each channel type. The classification of ion channels based on functional characteristics is largely supported by the sequence analysis of cloned channels and existing structural studies. Gating and Selective Impact Analysis: Conventional voltage clamping methods are usually used in the early stages of ion channel drug discovery and assay development to characterize the factors that control channel gating and determine ion selectivity. This information can then be used to design higher-density assays to provide higher throughput at the expense of reduced flexibility, control, and resolution.

Fluorescence Measurement: Fluorescence resonance energy transfer (FRET) is used by using a pair of dyes, phospholipid-anchored coumarin and hydrophobic oxanol (according to the rapid redistribution of the transmembrane field in the membrane), which can provide robustness and stability when studying voltage activity Reproducible signals, gated sodium and potassium channels. In fact, the assay can be adjusted to identify activators or inhibitors of a given ion channel.

Ion Flux Measurement: The development of indicators based on ion-selective fluorescent dyes has completely changed the method of ion flux measurement. These indicators improve the ease of use and temporal and spatial resolution of ion flux measurement. Genetically encoded ion flux indicators include chemiluminescent calcium sensors, aequorin and a variety of engineered fluorescent proteins are used continuously. The ion flux measurement method based on automatic flame photometry has been proved to be an effective alternative to some ion channel measurement methods.

High-throughput Screening Analysis: Ion channels are ideal targets for a wide range of health conditions. Traditional screening methods are labor-intensive and low-yield methods. Rapid advances in the development of functional tests and instruments have enabled high-throughput screening (HTS) activities to be realized in an ever-expanding list of channel types. However, many parameters need to be considered when optimizing HTS analysis, such as cell growth and harvest parameters, cell plating conditions, the type of fluorescent dye used for cell-based ion flux analysis, the type of quencher, and other parameters.
